In-vitro somatic embryogenesis from nucellar callus of mango (Mangifera indica L.).

Abstract : Immature fruitlets of mango cultivars Amrapali, Pusa Arunima, Kurukkan and Olour were collected at 25-30, 35-40 and 45-50 days after pollination for us in in vitro regeneration using nucellus. Maximum callus initiation was recorded for Olour (74.00%), followed by Kurukkan (70.10%). Callus induction was significantly affected by media composition; it was enhanced when 2,4-D alone was added to the culture medium. Nucellus from 35-45 day old fruitlets showed maximum callus induction. In terms of callus production, Kurukkan was the best genotype. The time taken for somatic embryo production was longest in Amrapali, followed by Pusa Arunima, Kurukkan and Olour. Amrapali also showed the highest frequency of somatic embryogenesis.
