Factors affecting in vitro somatic embryogenesis from nucellar tissues of Alphonso mango.

Abstract : Nucellar explants excised from fertilized ovules (1.0-2.1 cm long) of immature mango (cv. Alphonso) fruits (2.5-4.5 cm long) were cultured in Murashige and Skoog's (MS) medium or modified (1/2 strength) MS medium supplemented with 2,4-D (0.5, 1.0, and 2.0 g/litre) or 2,4-D + BAP [benzyladenine] (1.0 + 1.0, 2.0 + 1.0, or 2.0 + 2.0 mg/litre). To evaluate the effects of growth regulators on callus induction and somatic embryo formation, nucellar explants were also cultured in modified MS medium containing 2,4-D (1.0 or 2.0 mg/litre) with NAA, BAP, kinetin, or gibberellic acid (0.1, 0.5, or 1.0 mg/litre). The cultures were incubated at 25±2°C and 16 h photoperiod, and the growth parameters were observed after 45 days. Ovules 1.3 to 1.7 m long without zygotic embryos 0.3 to 0.5 cm long were most effective for embryogenic callus induction. The modified MS medium containing 400 mg glutamine, 100 mg ascorbic acid, 500 mg polyvinylpyrrolidone, 60 g sucrose, 1 mg 2,4-D, and 2.5 g phytagel/litre was superior for callus induction. The development and maturation of somatic embryos occurred in the same medium without 2,4-D.
