Cloning of Alphonso mango through vegetative explants.

Abstract : The effects of explant type (apical and axillary buds), age (mature tree explant, fresh sprout, and graft explant), and size (0.5, 1.0, 2.0. 3.0, and 5.0 cm), and of various antibrowning treatments (dipping of explants in 100 mg ascorbic acid + 50 mg citric acid for 30 seconds, dipping of explants in 0.5% PVP [polyvidone] + 3% sucrose for 30 seconds, incorporation of 0.05% PVP into the culture media, treatment 1 + treatment 3, and serial subculturing after 1 day) on the micropropagation of mango cv. Alphonso were studied. Culture establishment was most pronounced when apical buds (5 cm long) obtained from mature trees were used as explants. Dipping of explants in 100 mg ascorbic acid + 50 mg citric acid/100 ml distilled deionized water for 30 seconds, along with the incorporation of 0.05% PVP into the culture medium, was most effective in the reduction of browning incidence.
