Cloning and sequence analysis of MiLCYB2 gene promoter from mango (Mangifera indica L.).

Abstract : Objective: To study the expression and regulation rule of MILCYB2 in mango. Methods: A 991 bp fragment of MiLCYB2 promoter was cloned by chromosome walking method. Results: Promoter sequence analyzed by PlantCARE and PLACE showed that this promoter fragment had TATA-BOX, CAAT-BOX and some other regulatory elements, such as a variety of plant hormone cis-acting element involved in the abscisic acid responsiveness, the gibberellic acid responsiveness, the auxin responsiveness, the salicylic acid responsiveness and the MeJA responsiveness; and also cis-acting element was involved in circadian control, MYBHvl binding site and light responsive element. It was speculated that the expression of MILCYB2 was regulated by a variety of plant hormone, light, circadian, MYBHv1 transcription factors et al. Conclusion: Isolation and sequence analysis of MiLCYB2 promoter will help further explore the regulation of carotenoid biosynthesis pathway in mango.
