Molecular characterization of mango (Mangifera indica L.) using rapd markers.

Abstract : Genomic DNA was isolated from 90 days old leaves using C-TAB method. The purified DNA was amplified in PCR with RAPD primers. The banding pattern was recorded in the form of 0-1 sheet and was analyzed using UPGMA based Jaccard's similarity coefficient. The results were obtained in the form of dendrogram from RAPD analysis. Ten genotypes were grouped in two main clusters. Cluster II was the largest and included 7 genotypes while clusters I consists of 3 genotypes. The PIC value for the RAPD loci ranged from 0.00 to 0.99. Out of 8 RAPD primers, four generated polymorphic bands. A total of 27 bands were obtained, of which all were polymorphic, with an average of 7 bands per primer. On the basis of PIC value RAPD primers OPA 10, OPD 7, OPA 13 and OPC 12 provide maximum accessions coverage in the mango genome. These primers were found of high importance for characterization of mango genotypes.
